The basic principle is to combine an antigen or antibody with an enzyme to form an enzyme-labeled antigen or antibody. The degree of color change can be used to semi-quantitatively or quantitatively detect antigens or antibodies in the sample.

Indirect ELISA kits are mainly used for detecting antigens, while direct ELISA kits are mainly used for detecting antibodies.
