      The original antibodies are susceptible to degradation in vivo by factors such as proteases, oxidants, and heat. By introducing modified groups, we can enhance the stability of the antibodies and prolong their half-life in the body, thereby increasing their therapeutic efficacy and persistence. These states of protein expression and activation include phosphorylation, acetylation, glycosylation, prenylation, myristoylation, ubiquitination, sumoylation, protein cleavage (neo-epitopes), splice variants, and isoforms. 

      However, the process of producing antibodies against a phospho-residue is more complicated than using peptide immunogens. Phospho-specific antibodies are generated using peptides containing one or more phosphorylated amino acids. There are three residues which can be phosphorylated: Serine (S), Threonine (T) and Tyrosine (Y). 

      BOT Bioscience provides a custom phospho-specific antibody production service. Typically such a project is a monoclonal anti-peptide antibody. We can use the Phospho-Peptides to immunize the mice, and use Phospho-Peptides to screen for positive hybridoma clones, and use non-phospho-peptides to do negative selection. This negative selection is required since peptide phosphorylation or protein phosphorylation is never 100% complete.
